L-arginine serves as a precursor for nitric oxide (NO) synthesis, a molecule that promotes vasodilation — the widening of blood vessels. When endothelial cells convert L-arginine into nitric oxide via the enzyme nitric oxide synthase (NOS), this NO activates cyclic guanosine monophosphate (cGMP) within smooth muscle tissue, causing the relaxation of penile arteries and corpus cavernosum muscles, thereby enabling erection.
